CRISPR-Cas9 Project
CRISPR-Cas9 is a gene editing technology found in some bacteria. Through a hands-on activity and an online simulation, we have been learning about CCASPR-Cas9 and its function in the human body.
CRISPR is an acronym for clustered regularly interspaced short palindromic repeats. CRISPR-cas9’s structure consists of a guide RNA strand that acts like a pair of scissors inside the Cas-9 nuclease.
In targeting the Cas9 enzymes searches for the target DNA, when it recognizes the PAM site binding begins. Cas9 binds to the PAM site. The PAM site is made of any nucleotide followed by two guanines. This sequence is rich in human genomes. After binding the Cas9 unwinds the DNA double helix at a point above the PAM site.
If the strand is not an exact match to the 20-nucleotide within the guide RNA, CRISPR-Cas9 disengages from the DNA and the DNA returns to its double helix shape. If they are a match the guide RNA complimentary base pairs to the DNA strand forming a new helix. The binding event activates the DNA cutting. The cutting happens specifically at a point 3 nucleotides above the PAM site. Cas9 generates the cut and cleaves both the DNA and RNA strand resulting in a DNA break.
DNA can be repaired in 2 different ways nonhomologous ends joining (NHEJ) or homology-directed repair (HDR). NHEJ is more commonly used, it is faster as it does not use a template for the repair by joining the two broken ends together. NHEJ is an error prone process that can introduce a new mutation. HDR is a less error prone process, it uses a DNA template to accurately repair the break. When the break is repaired Cas9 will once again cut it with the same process. Scientists use CRISPR-Cas9 to modify and target our DNA sequence to remove mutations which benefits human lives.
The modelling activity accurately demonstrated how the CRISPR-cas9 attaches to the DNA strand. This activity misrepresents how the DNA is cut down the middle. It would be more effective if there was a way to demonstrate the cleaving process clearly.
I do believe that models are an effective way of learning. Models are a great visual learning activity as it helps to see the bigger picture that words on a paper can’t show. Models are often a way to be hands on that is also an effective way to learn.

The Eddy Article – “Riverside prepares for this year’s school apparel”
Riverside prepares for this year’s school apparel
Riverside’s Marketing and Promotion class taught by Mr. Rothenberger, will be releasing a variety of new school merchandise.
by Coral Ford
Mr. Rothenberger’s Marketing and Promotion class is researching and brainstorming merchandise for the 2021-2022 Riverside Secondary school year.
“The process all started with a grand vision of having a place where Riverside students could come to purchase school merch and other convenience items, and also have a place to hang out at lunch, etc.,” said Richard Rothenberger, Marketing and Promotion teacher at Riverside Secondary.
The lounge in room 224 already has stock, “you should come by and check it out anytime before period 2, at lunch, or after period 5. We have merch on display and ready for purchase, with another large order on the way,” said Rothenberger.
With the new year came new options. In addition to hoodies and crewnecks, they started selling camping blankets, wind breakers, toques, varsity jackets, and other miscellaneous items explained Rothenberger.
The lounge has been approved to use tap to pay making it ever easier for students to pay.
The lounge is adamant about staying open all year long with popular items available to purchase. Although there will be a schoolwide online order system, so that parents and students can pay remotely.
“We would like to help any group within the school that is looking to provide product or raise funds by making our space available as a hub for all purchases in the school,” said Rothenberger.
The students have set up an account on Instagram @theriversidelounge_ with updates on new products, promotion, giveaways, and to provide feedback Rothenberger shared. There will also be advertising on rapidevents, riversideathletics, and on the Riverside web page.
“Our hope is that students will automatically think of the Lounge when they are looking to purchase something,” said Rothenberger.
“At Riverside, the WAVE was originally our school store… thanks to Mr. Bruneau and his support we are now able to re-open the school store and help other departments throughout the school with merchandise sales,” said Rothenberger.
Indigenous Podcast – Hitch and Run
Barbara Kentner is a woman whose death inspired the Thunder Bay indigenous community to stand up and demand a voice. Around her legacy we created a podcast entitled, “Hitch and Run” that share’s the story of Barbara Kentner and the effects it had on the community. To create this podcast, we researched the life of Kentner before the incident, during her hospital time, and her family and community after her passing. Then we integrated the information into a script. Then we used a Blue Snowball Microphone, provided by Vancity. During the proses we made a cover image for the podcast. We took inspiration from the Barbara Kentner billboard. We used Audacity to record, edit, and incorporate royalty free music to set the tone of the podcast.
